Effects of Dietary Xylooligosaccharide on Antioxidative Defense System of Liver and Changes of Hepatocytes in Rat Fed High Cholesterol Diets

Abstract
This study was conducted to examine the effects of dietary xylooligosaccharide on antioxidative defense system of liver and changes of hepatocytes in rat fed high cholestrol diet. Sprague-Dawley male rats weighing 100¡¾10 g were randomly divided into group of one normal diet, and four high cholesterol diet containing 1% cholesterol. The high cholesterol(1%) diet groups were classified into xylooligosaccharide free diet(CXO group), 5% xylooligosaccharide diet(C5XO group), 10% xylooligosaccharide diet(C10XO group) and 15% xylooligosaccharide diet(C15XO group) according to the levels of dietary xylooligosaccharide supplementation. The experimental diets were fed ad libitum for 4 weeks. The weights of liver were significantly higher than those of normal and CXO groups. Activities of superoxide dismutse(SOD) and glutathione peroxidase(GSHpx) in liver were significant lower in CXO group than in normal group but those of xylooligosaccharide groups were significantly higher than in CXO group. C10XO group was the most effective in any other concentrations. The content of thiobarbituric acid reactive substance(TBARS) in liver was 211% higher in CXO group than nomal group. These of TBARS in C5XO, C10XO and C15XO groups were decreased 30%, 50% and 33%, respectively, compared with CXO group. Serum GOT was increased in CXO group but that of xylooligosaccharide groups was looking clear due to large amount of incracytoplasmic fat. Liver of fatty change in C10XO group is quite diminished compared with CXO group. Thus, dietary xylooligosaccharide supplemention play an important role in favorable changes in morphaologies of liver an serves as antioxidant against lipid peroxidation in rat fed high cholesterol diet. These results suggest that xylooligosaccharides are potent in lowering liver lipids.
